STAR editors comprise lgRNA constructs with self-contained DNA repair templates for precise gene editing and require no additional donor DNA. In addition, lgRNAs exclude certain inactive secondary structures of sgRNAs and thus are more active and potentially decrease drug dosages and related adverse effects. STAR editors are delivered as RNP
conjugates or as RNA mixtures in delivery carriers.
LgRNA technology: LgRNAs are intellectually distinct next-generation single-molecule CRISPR guide RNAs with improved drug properties (US10059940B2). Guide RNA directs where the enzyme cuts and is crucial in precise gene editing. Chemical modifications of guide RNAs are required for better editing efficacy and selectivity, targeted delivery and decreased toxicity and can only be introduced by chemical synthesis.
